| Description | テラピアコラーゲンの持つ間葉系幹細胞の骨芽細胞への分化誘導効果を、トランスグルタミナーゼによる架橋をコラーゲンに施すことにより、長期間持続できることが分かった。 Abstract: In this study, fish collagen derived from tilapia scales which are free from the risk of infection was employed to fabricate a 3D porous scaffold. Tilapia scale collagen (TSC) scaffolds were modified using microbial transglutaminase (mTGase) enzyme as a catalyst to preserve the inherent properties of collagen material. Human mesenchymal stem cells (hMSCs) were incubated with various substitutes including TSC and porcine collagen-coated cell culture dishes with or without mTGase, TSC and porcine collagen scaffolds crosslinked by mTGase. Differentiation of hMSCs was verified using alkaline phosphatase activity and osteocalcin assays. A remarkably enhanced expression of osteoblastic differentiation was observed selectively with TSC-coated dishes with/without mTGase and TSC collagen scaffold crosslinked by mTGase. Thus, the combination of TSC and the mTGase crosslinking method will be useful for osteoblastic differentiation and bone tissue engineering. | |||||||||||||||
